DNA methylation Markers in the Detection of Hepatocellular Carcinoma

چکیده
Hepatocellular carcinoma (HCC) is the most common primary liver malignancy and has a poor prognosis. Epigenetic modification been shown to be deregulated during HCC development by dramatically impacting differentiation, proliferation function of cells. One important epigenetic DNA methylation which methyl groups are added cytosines without changing sequence itself. Studies found that methylated markers can specific for detection HCC. On basis these findings utility as novel biomarkers early-stage measured in blood, indeed superior sensitivity specificity have several studies when compared current surveillance methods. However, variety factors currently limit immediate application exciting biomarkers. In this review, we provide detailed rationalization approach use detection, summarize recent on focusing importance etiological cause disease mechanisms leading cancer.

DNA methylation of tumor suppressor genes in hepatocellular carcinoma
The basic unit of chromatin is a nucleosome included an octamer of the four core histones and 147 base pairs of DNA. Posttranslational histones modifications affect chromatin structure resulting in gene expression changes. CpG islands hypermethylation within the gene promoter regions and the deacetylation of histone proteins are the most common epigenetic modifications. The aberrant patterns of...

Hepatocellular carcinoma (HCC) is the 5th commonest malignancy worldwide and is the third most common cause of cancer-related death. The prevalence is different in the world. The ability to predict patients at higher risk of recurrence and with a poor prognosis would help to guide surgical and chemotherapeutic treatment according to individual risk. As understanding of hepatocarcinogenesi...
متن کاملElucidating the Landscape of Aberrant DNA Methylation in Hepatocellular Carcinoma
BACKGROUND Hepatocellular carcinoma (HCC) is one of the most common cancers and frequently presents with an advanced disease at diagnosis. There is only limited knowledge of genome-scale methylation changes in HCC. METHODS AND FINDINGS We performed genome-wide methylation profiling in a total of 47 samples including 27 HCC and 20 adjacent normal liver tissues using the Illumina HumanMethylati...
متن کاملDNA methylation markers and serum α-fetoprotein level are prognostic factors in hepatocellular carcinoma.
INTRODUCTION Hypermethylation of relevant genes may affect the prognosis of patients with cancer. The purpose of this study was to analyze whether methylation of the promoter regions of cell cycle regulators as well as elevated α-Fetoprotein (AFP) levels are useful prognostic factors for patients with hepatocellular carcinoma (HCC). MATERIAL AND METHODS Nested methylation-specific PCR (nested...
متن کاملPharmacological Unmasking Microarray Approach-Based Discovery of Novel DNA Methylation Markers for Hepatocellular Carcinoma
DNA methylation is one of the main epigenetic mechanisms and hypermethylation of CpG islands at tumor suppressor genes switches off these genes. To find novel DNA methylation markers in hepatocellular carcinoma (HCC), we performed pharmacological unmasking (treatment with 5-aza-2'-deoxycytidine or trichostatin A) followed by microarray analysis in HCC cell lines. Of the 239 promoter CpG island ...
